you should be fine. it doesn't matter where the mail routes through. what matters is where the !besadmin and user mailboxes reside.
bes 4.1 is not compatible with e2010 so before u migrate anyone u'll need to upgrade to bes 5.0 or later, the migrate the !besadmin account.
