With Key Lock engaged, your keys will not do anything, but if depressed, the backlight will power. This will not occur when in Standby. I actually use both. I have an application that Autolocks my keyboard after the backlight goes out, and if I'm going to put my phone in a pocket, I also engage Standby so the backlight won't come on and wear down the battery.

I use keyboard lock, and never standby. I have my right convenience key set to keyboard lock. Reason for this: I'm frequently listening to music on my BB and want to put the phone otherwise "to sleep" while I keep listening to my music. The convenient standby button on top is also the mute button. It won't cooperate (go to standby) if you're playing music at the time.
I don't use a magnetized holster, so I have no clue whether if you put a BB playing music into one of these and it enters standby, the music will stop. But I am certain that pressing the top button to put your phone to sleep will make any music you are playing stop, and that using keyboard lock will not.

Actually if you are playing music, you can still put the phone to sleep. Make sure you are out of the media player. Press the standby button once to pause the music, press it a second time and the music should continue and put the device into standby mode. If you have OS4.5 I think you can even move back/forward through the tracks using the side volume buttons.
